Single stranded DNA-fragments containing short runs of isoguanine such as d(T(4)iG(4)T(4)) (5) or d(iG(4)T(4)) (6) form quartet structures by self-assembly of the isoguanine residues. The stoichiometry of the complexes is deduced from mixed aggregates formed between d(T(4)iG(4)T(4)) and d(iG(4)T(4)). The iG(d)-tetrads are more stable with regard to their thermal denaturation and to their resistance against enzymatic phosphodiester hydrolysis than those formed by dG.
